Transcriptome analysis and molecular mechanism of linseed (Linum usitatissimum L.) drought tolerance under repeated drought using single-molecule long-read sequencing

Abstract Background Oil flax (linseed, Linum usitatissimum L.) is one of the most important oil crops., However, increases in drought resulting from climate change have dramatically reduces linseed yield and quality, but very little known about how coordinates expression resistance gene response to different level stress (DS) on genome-wide level. Results To explore transcriptional DS repeated (RD) stress, we determined tolerance varieties. Then performed full-length transcriptome sequencing drought-resistant variety (Z141) drought-sensitive (NY-17) under RD at seedling stage using single-molecule real-time RNA-sequencing. Gene Ontology (GO) reduce visualize GO (REVIGO) enrichment analysis showed that upregulated genes Z141 were enriched more functional pathways related plant than those NY-17 DS. In addition, 4436 transcription factors identified, 1190 responsive treatments. Moreover, protein-protein interaction (PPI) network proline biosynthesis pathway interacts with through RAD50 (DNA repair protein 50) interacting 1 (RIN-1). Finally, DNA structural patterns verified by RT- PCR. Conclusions The may be its upregulation Proline play an role maintaining cell osmotic protecting ROS damage. summary, this study provides a new perspective understand adaptability linseed.
